Multiple-dose, 6-day, 3-way crossover study in healthy subjects (n=14) compared mean plasma VPA levels of Depakote ER 1000 mg qd fasting to Depakote 500 mg q12h fasting. Depakote ER given once-daily produced an average bioavailability of 89% relative to an equal total daily dose of Depakote given BID. The median time to maximum plasma valproate concentrations (Cmax) after Depakote ER administration was 17 hours.
